RESUMO

This study evaluated the effects of glycoelectrolytic supplements on the performance, blood parameters, and intestinal morphology of piglets during the post-weaning period. In the study, a total of 240 piglets weaned aged 17 22.60 + 1.10 days were used. The control group (n = 120) received only water, and the treatment group (n = 120) received an oral glycoelectrolytic supplement diluted in water (0.75%) during the first three days after weaning. Feed intake, daily weight gain, final weight, feed conversion ratio, and post-weaning mortality were evaluated. On the third day after weaning, the blood glucose levels of all piglets were analyzed. Blood was collected from 12 piglets from each treatment group on day 3 after weaning for blood count analysis, and intestinal fragments were collected for anatomopathological and morphometric evaluation. Better feed conversion ratio (1.29) and higher consumption of liquids (0.639 L/day) were observed in the piglet group supplemented with glycoelectrolytes on day 3 after weaning (P < 0.05). The supplemented group presented a higher glycemic index (80.78 mg/dL), average corpuscular volume (67.35 fL), and average corpuscular hemoglobin (20.46 pg) than the control group (P < 0.05). The evaluation of intestinal integrity and the probability of diarrhea occurrence were similar between the groups (P > 0.05). Oral glycoelectrolytic supplementation can be an option for piglets immediately after weaning as it improves feed conversion and consumption of liquids, in addition to increasing blood glucose without the occurrence of diarrhea, thus reducing dehydration and energy deficit.

The use of blood metabolites (BM), fecal starch (FS), and apparent digestion of starch, (ATTSD) as indicators of feed efficiency (FE) in beef cattle in the feedlot was studied. Fourteen bulls were used, originating in an industrial cross, without a defined racial group, with mean body weight of 284.86kg, individually fed, being evaluated in a 42-day confinement system. After the evaluation, the animals were divided into two groups according to the individual FE: high feed efficiency (HE) and low feed efficiency (LE). There was a difference between the groups in the variables FE, feed conversion (FC), final weight (FW), and daily weight gain (DWG). The FE had a positive correlation with DWG, FC, and FW. There was no difference between the groups for the variables BM, FS, and ATTSD, nor was there any correlation between these variables and FE. Considering the feed cost, the HE animals proved more profitable. BM, FS, and ATTSD did not statistically show potential to be used as indicators of FE, despite the evidence of numerical differences of these variables between the different groups, tendency of correlations with FE, and discriminating function with potential assertiveness.(AU)


Foi estudada a utilização dos metabólitos sanguíneos (BM), do amido fecal (FS) e da digestão aparente do amido (ATTSD) como indicadores de eficiência alimentar (FE) em bovinos de corte em confinamento. Utilizaram-se 14 touros, originários de cruzamento industrial, sem grupo racial definido, peso corporal médio de 284,86kg, alimentados individualmente, sendo avaliados em sistema de confinamento por 42 dias. Após a avaliação, dividiram-se os animais em dois grupos, de acordo com a FE individual: alta eficiência alimentar (HE) e baixa eficiência alimentar (LE). Houve diferença entre os grupos nas variáveis FE, conversão alimentar (FC), peso final (FW) e ganho de peso diário (DWG). A FE teve correlação positiva com DWG, FC e FW. Não houve diferença entre os grupos para as variáveis BM, FS e ATTSD, tampouco houve correlação entre essas variáveis e a FE. Considerando-se o custo alimentar, os animais HE mostraram-se mais lucrativos. BM, FS e ATTSD não mostraram, estatisticamente, potencial para serem utilizados como indicadores de FE, apesar da evidência de diferenças numéricas dessas variáveis entre os diferentes grupos, tendência de correlações com a FE e de função discriminante com potencial assertividade.(AU)

The aim of this study was to evaluate swine females of different genetic lines submitted to different reproductive management and housing systems during pregnancy on reproductive performance and animal welfare parameters. After artificial insemination protocol, 524 females were divided into two gestation housing systems: PEN1=animals housed in individual stalls during the breeding and after group-housed; PEN32=animals housed in individual stalls from breeding until 32 days of pregnancy and after group-housed. The number of piglets born, and the pregnancy and farrowing rates were evaluated. Welfare parameters related to the pregnancy phase were used. Females who weaned more piglets in the previous farrowing had a higher number of piglets born at the next farrowing. The pregnancy rate was affected by the number of semen doses. The farrowing rate was not influenced by the evaluated parameters, with average value of 91.36%. There was no effect of the gestation housing system and the genetic lines on pregnancy and farrowing rates, with values above 90.0%. The animal welfare indicators showed more compromised parameters in PEN1 system. PEN1 system did not impair the reproductive performance although it presented more compromised animal welfare parameters.(AU)


O objetivo deste estudo foi avaliar fêmeas suínas de diferentes linhagens genéticas submetidas a diferentes sistemas reprodutivos de manejo e alojamento durante a prenhez sobre parâmetros de desempenho reprodutivo e bem-estar animal. Após o protocolo de inseminação artificial, 524 fêmeas foram divididas em dois sistemas de alojamento de gestação: PEN1=animais alojados em baias individuais durante o protocolo de inseminação artificial e, depois, alojados em grupo; PEN32=animais alojados em baias individuais desde o protocolo de inseminação artificial até 32 dias de prenhez e, depois, alojados em grupo. O número de leitões nascidos e as taxas de prenhez e parto foram avaliados. Parâmetros de bem-estar relacionados à fase gestacional foram utilizados. As fêmeas que desmamaram mais leitões no parto anterior tiveram um maior número de leitões nascidos no próximo parto. A taxa de prenhez foi afetada pelo número de doses de sêmen. A taxa de parto não foi influenciada pelos fatores avaliados, com valor médio de 91,36%. Não houve efeito do sistema de alojamento gestacional e das linhas genéticas sobre as taxas de prenhez e parto, com valores acima de 90,0%. Os indicadores de bem-estar animal mostraram parâmetros mais comprometidos no sistema PEN1. O sistema PEN1 não prejudicou o desempenho reprodutivo, embora tenha apresentado parâmetros de bem-estar animal mais comprometidos.(AU)

A hipótese deste estudo é de que o uso da combinação de butafosfan e cianocobalamina pode melhorar a resistência periférica à insulina, aumentar a quantidade de glicose disponível para a glândula mamária e a produção de leite. Assim, o objetivo foi investigar os efeitos combinados de butafosfan e cianocobalamina sobre o metabolismo da glicose em vacas leiteiras no período pós-parto. Vinte e uma vacas leiteiras foram divididas em dois grupos: grupo controle (CON, n= 11), que recebeu cinco aplicações de solução salina (20mL / animal 0,9% NaCl), e grupo Catosal(r) (ABC, n= 10), que recebeu cinco aplicações de 20mL de uma solução contendo as substâncias butafosfan e cianocobalamina (B12 Catosal(r), 100mg da substância butafosfan e 50µg de cianocobalamina por mL). As aplicações foram realizadas por via intramuscular, nos dias sete, 12, 17, 22 e 27 pós-parto. As amostras de sangue foram coletadas para a avaliação das concentrações plasmáticas de fósforo, glicose, ácidos graxos não esterificados (AGNE), albumina, aspartato aminotransferase (AST) e creatina quinase (CK). Nos dias oito e 28 pós-parto, os animais foram pesados e submetidos aos testes de tolerância à glicose e à insulina. O tratamento promoveu perda de peso (ABC 40,4kg, CON 10,73kg, P<0,05) e aumento da AST (ABC 62,92 ±3,31U/L, CON 53,11±3,49 U / L, P<0,05) e dos níveis de CK (ABC 134,09± 19,08U / L, CON 79,43 ± 18,27U / L). Os grupos não diferiram quanto ao metabolismo (área sob a curva) da glicose nos dias oito e 28, porém os animais tratados tiveram um aumento na glicemia (P<0,05) no dia 28 pós-parto (97,54 ± 8,54mg / dL), após a administração de insulina, em comparação ao dia oito (83,01 ± 8,54mg / dL). Assim, pode-se concluir que a combinação de butafosfan e cianocobalamina melhora a adaptação do metabolismo da glicose em vacas leiteiras no início da lactação.(AU)


The hypothesis of this study is that the combined use of butaphosphan and cyanocobalamin could enhance peripheral insulin resistance, increasing the amount of glucose available for the mammary gland and milk production. Thus, our aim was to investigate the combined effects of butaphosphan and cyanocobalamin on the glucose metabolism in dairy cows during the postpartum period. Twenty one dairy cows were divided into two groups: Control Group (CON, n= 11), that received 5 injections of saline solution (20mL/animal 0.9 % NaCl), and Catosal group (ABC, n= 10) which received 5 injections of 20mL of a Butafosfan and Cyanocobalamin solution (Catosal(r) B12, 100mg of Butafosfan and 50µg Cyanocobalamin for mL). The injections were performed by intramuscular route, on days 7, 12, 17, 22 and 27 postpartum. Blood samples were taken on these days to evaluate plasma concentrations of phosphorus, glucose, non-esterified fatty acids (NEFA), albumin, aspartate aminotransferase (AST) and creatine kinase (CK). On days 8 and 28 postpartum, the animals were weighted and subjected to the glucose tolerance and insulin challenge tests. The treatment promoted weight loss (ABC 40.4kg, CON 10.73kg, P< 0.05) and increased AST (ABC 62.92 ±3.31U/L, CON 53.11 ±3.49U/L, P< 0.05) and CK levels (ABC 134.09 ±19.08U/L, CON 79.43 ±18.27U/L). Glucose metabolism (area under the curve) did not differ (P> 0.05) among groups on days 8 and 28; however, ABC animals showed higher serum glucose levels (P< 0.05) after insulin administration on day 28 postpartum (97.54 ±8.54mg/dL) when compared to day 8 (83.01 ±8.54mg/dL). It could be concluded that the combined use of butaphosphan and cyanocobalamin interferes positively with the adaptation of glucose metabolism in dairy cows in early lactation.(AU)

Compararam-se as concentrações de cálcio, glicose e insulina em ovelhas gestantes e não gestantes submetidas ao teste de tolerância à glicose (TTG). Oito ovelhas gestantes (GG) e oito não gestantes (NG) foram submetidas ao TTG por meio da administração endovenosa de uma solução de glicose, na dose de 500mg/kg de peso vivo. Amostras de sangue foram coletadas nos tempos de -15, zero, 15, 30, 45, 60, 90, 120, 150 e 180 minutos, sendo zero min o momento da infusão, para avaliação da concentração sanguínea de glicose, cálcio e insulina. Foi avaliada a taxa de metabolização de glicose e insulina, por meio do cálculo da área sob a curva (ASC). As concentrações de glicose, cálcio, insulina e a ASC da insulina não diferiram entre os grupos (P>0,05), entretanto as três últimas ASCs da glicose foram mais elevados nas gestantes (P<0,05), que correspondem ao período entre 90 e 180min. O teste de correlação de Pearson revelou correlação negativa entre concentração de cálcio e insulina nas gestantes (P<0,05). Estes resultados indicam menor taxa de metabolização de glicose em ovelhas gestantes e uma possível relação de cálcio e insulina no terço final de gestação.


The aim of this study was to investigate glucose, insulin and calcium metabolism during the Glucose Tolerance Test (GTT) in pregnant or nonpregnant ewes. Eight pregnant (PG) and nonpregnant (NPG) ewes per group received a glucose endovenosus bolus containning 500mg/kg in one minute. Blood samples were collected on -15, 0, 15, 30, 45, 60, 90, 120, 150 and 180 minutes interval (0 min - injection). These samples were used to evaluate glucose, calcium, insulin concentration, as well as insulin and glucose metabolization rate, which were determined by calculating the Area Under the Curve (AUC). The calcium, glucose and insulin concentrations were not different between groups. However, the three last glucose ASCs were higher in the pregnant group and in this group calcium and insulin blood concentrations were negatively correlated .These results showed lower glucose tolerance in pregnant ewes and suggest a link between insulin and calcium in the late pregnancy.

We present a study of the infrared (IR) spectra of the (0001) deuterated ice surface based on first-principles molecular dynamics simulations. The computed spectra show a good agreement with available experimental IR measurements. We identified the bonding configurations associated with specific features in the spectra, allowing us to provide a detailed interpretation of IR signals. We computed the spectra of several proton ordered and disordered models of the (0001) surface of ice, and we found that IR spectra do not appear to be a sensitive probe of the microscopic arrangement of protons at ice surfaces.

The aim of this study was to determine the effect of prepartum somatotropin injection in late-pregnant Holstein heifers on metabolism, milk production and resumption of postpartum ovulation. For this study, 31 late-pregnant Holstein heifers were used. The heifers were assigned randomly into two treatments: (1) 500 mg sc injections of somatotropin (somatotropin treatment, n = 15) at -35 and -21 days, and, if pertinent, at -7 days from expected calving date and (2) no treatment (control group, n = 16). Blood samples were collected weekly from -5 to 7 weeks after calving. Heifers with progesterone concentrations in plasma above 1 ng/ml in two consecutive postpartum samples were considered as having resumed ovarian activity. A higher proportion (P = 0.04) of heifers treated with somatotropin resumed ovarian activity in the first 7 weeks post partum (73.3%; 11/15) compared with the control group (37.5%; 6/16). A higher number (P = 0.02) of heifers in the somatotropin treatment group also ovulated during the first postpartum follicular wave (53.3%; 8/15) compared with the control group (12.5%; 2/16), as indicated by the number of heifers ovulating in the first 3 weeks post partum. Pregnancy rate was not affected by treatments (P > 0.10) and averaged 40.0% (6/15) in somatotropin-treated and 25.0% (4/16) in control heifers when evaluated up to 150 days in milk. Somatotropin treatment increased the average daily milk production by 2.8 kg/cow per day (P < 0.0001) and reduced the somatic cell count (P = 0.009). Plasma IGF-I was higher (P < 0.05) for somatotropin-treated heifers in the prepartum period. Insulin and body condition score were higher (P < 0.05) and non-esterified fatty acids were lower (P < 0.05) for somatotropin-treated cows in the early postpartum period. In conclusion, somatotropin injection during the prepartum period in late-pregnant Holstein heifers was able to increase the proportion of heifers resuming ovarian activity early post partum, inspite of higher milk production.

RESUMO

We present density-functional theory calculations of the optical absorption spectra of silica glass for temperatures up to 2400 K. The calculated spectra exhibit exponential tails near the fundamental absorption edge that follow the Urbach rule in good agreement with experiments. We discuss the accuracy of our results by comparing to hybrid exchange correlation functionals. We show that the Urbach rule holds in a frequency interval where optical absorption is Poisson distributed with very large statistical fluctuations. In this regime, a direct relation between the optical absorption coefficient and electronic density of states is derived, which provides a link between photoemission and absorption spectra and is used to determine the lower bound to the Urbach frequency regime.

We report on the use of first-principles molecular dynamics calculations to examine properties of liquid carbon dioxide in the pressure-temperature range of 0-1 TPa and 200-100 000 K. The computed equations of state points are used to predict a series of shock Hugoniots with initial starting conditions that are relevant to existing and ongoing shock-wave experiments. A comparison with published measurements up to 70 GPa shows excellent agreement. We find that the liquid undergoes a gradual phase transition along the Hugoniot and have characterized this transition based on changes in bonding and structural properties as well as the conductivity and reflectivity of the fluid.

Ebola virus-like particles (VLPs) were produced in insect cells using a recombinant baculovirus expression system and their efficacy for protection against Ebola virus infection was investigated. Two immunizations with 50 microg Ebola VLPs (high dose) induced a high level of antibodies against Ebola GP that exhibited strong neutralizing activity against GP-mediated virus infection and conferred complete protection of vaccinated mice against lethal challenge by a high dose of mouse-adapted Ebola virus. In contrast, two immunizations with 10 microg Ebola VLPs (low dose) induced 5-fold lower levels of antibodies against GP and these mice were not protected against lethal Ebola virus challenge, similar to control mice that were immunized with 50 microg SIV Gag VLPs. However, the antibody responses against GP were boosted significantly after a third immunization with 10 microg Ebola VLPs to similar levels as those induced by two immunizations with 50 microg Ebola VLPs, and vaccinated mice were also effectively protected against lethal Ebola virus challenge. Furthermore, serum viremia levels in protected mice were either below the level of detection or significantly lower compared to the viremia levels in control mice. These results show that effective protection can be achieved by immunization with Ebola VLPs produced in insect cells, which give high production yields, and lend further support to their development as an effective vaccine strategy against Ebola virus.

We evaluate the accuracy of density functional theory quantum calculations of biomolecular subsystems using a simple electrostatic embedding scheme. Our scheme is based on dividing the system of interest into a primary and secondary subsystem. A finite difference discretization of the Kohn-Sham equations is used for the primary subsystem, while its electrostatic environment is modeled with a simple one-electron potential. Force-field atomic partial charges are used to generate smeared Gaussian charge densities and to model the secondary subsystem. We illustrate the utility of this approach with calculations of truncated dipeptide chains. We analyze quantitatively the accuracy of this approach by calculating atomic forces and comparing results with full QM calculations. The impact of the choice made in terminating dangling bonds at the frontier of the QM region is also investigated.

RESUMO

We report on the first single-state measurement of the broadband (450-800 nm) dielectric function of gold isochorically heated by a femtosecond laser pulse to energy densities of 10(6) - 10(7) J/kg. A Drude and an interband component are clearly seen in the imaginary part of the dielectric function. The Drude component increases with energy density while the interband component shows both enhancement and redshift. This is in strong disagreement with predictions of a recent calculation of dielectric function based on limited Brillouin zone sampling.

The application of mass spectrometry to identify disease biomarkers in clinical fluids like serum using high throughput protein expression profiling continues to evolve as technology development, clinical study design, and bioinformatics improve. Previous protein expression profiling studies have offered needed insight into issues of technical reproducibility, instrument calibration, sample preparation, study design, and supervised bioinformatic data analysis. In this overview, new strategies to increase the utility of protein expression profiling for clinical biomarker assay development are discussed with an emphasis on utilizing differential lectin-based glycoprotein capture and targeted immunoassays. The carbohydrate binding specificities of different lectins offer a biological affinity approach that complements existing mass spectrometer capabilities and retains automated throughput options. Specific examples using serum samples from prostate cancer and hepatocellular carcinoma subjects are provided along with suggested experimental strategies for integration of lectin-based methods into clinical fluid expression profiling strategies. Our example workflow incorporates the necessity of early validation in biomarker discovery using an immunoaffinity-based targeted analytical approach that integrates well with upstream discovery technologies.

RESUMO

The increasing threat of bioterrorism and continued emergence of new infectious diseases has driven a major resurgence in biomedical research efforts to develop improved treatments, diagnostics and vaccines, as well as increase the fundamental understanding of the host immune response to infectious agents. The availability of multiple mass spectrometry platforms combined with multidimensional separation technologies and microbial genomic databases provides an unprecedented opportunity to develop these much needed resources. An overview of current proteomic strategies applied to microbes and viruses considered potential bioterrorism agents is presented. The emerging area of immunoproteomics as applied to the development of new vaccine targets is also summarized. These powerful research approaches can generate a multitude of potential new protein targets; however, translating these proteomic discoveries to useful counter-bioterrorism products will require large collaborative research efforts across multiple basic science and clinical disciplines. A translational proteomic research paradigm illustrating this approach using influenza virus as an example is discussed.

Proteomic profiling of serum is an emerging technique to identify new biomarkers indicative of disease severity and progression. The objective of our study was to assess the use of surface-enhanced laser desorption/ionization time-of-flight mass spectrometry (SELDI-TOF MS) to identify multiple serum protein biomarkers for detection of liver disease progression to hepatocellular carcinoma (HCC). A cohort of 170 serum samples obtained from subjects in the United States with no liver disease (n = 39), liver diseases not associated with cirrhosis (n = 36), cirrhosis (n = 38), or HCC (n = 57) were applied to metal affinity protein chips for protein profiling by SELDI-TOF MS. Across the four test groups, 38 differentially expressed proteins were used to generate multiple decision classification trees to distinguish the known disease states. Analysis of a subset of samples with only hepatitis C virus (HCV)-related disease was emphasized. The serum protein profiles of control patients were readily distinguished from each HCV-associated disease state. Two-way comparisons of chronic hepatitis C, HCV cirrhosis, or HCV-HCC versus healthy had a sensitivity/specificity range of 74% to 95%. For distinguishing chronic HCV from HCV-HCC, a sensitivity of 61% and a specificity of 76% were obtained. However, when the values of known serum markers alpha fetoprotein, des-gamma carboxyprothrombin, and GP73 were combined with the SELDI peak values, the sensitivity and specifity improved to 75% and 92%, respectively. In conclusion, SELDI-TOF MS serum profiling is able to distinguish HCC from liver disease before cirrhosis as well as cirrhosis, especially in patients with HCV infection compared with other etiologies.

The dissociation of water under pressure is investigated with a series of ab initio molecular dynamics simulations at thermodynamic conditions close to those obtained in shock wave experiments. We find that molecular dissociation occurs via a bimolecular process similar to ambient conditions, leading to the formation of short-lived hydronium ions. Up to twofold compression and 2000 K, the oxygen diffusion coefficient is characteristic of a fluid. Our findings do not support models used to estimate the liquid electrical conductivity and interpret Raman spectra that assume the presence of free protons.

Water under pressure is investigated by first principles molecular dynamics, with a focus on the changes in hydrogen bonding and the oxygen network in the nondissociative regime. At a pressure of 10 GPa and a temperature of 600 K, which is close to the freezing point, no appreciable molecular dissociation is observed in the simulations. However, the structure of water is substantially altered from that at ambient conditions. The liquid exhibits a much larger coordination of oxygen atoms, an essential weakening of hydrogen bonding, and sizable changes in the density of electronic states close to the Fermi level. Our results provide new structural data for direct comparison with future experiments.
